The Investigation of Cobalt Chloride effects on Some Toxicokinetic Parameters of Cyanide and Thiocyanate

This study was made to determine the some toxicokinetic parameters of eyanide and thiocyanate in sheep experimentally poisoned with amygdalin. In this study. 1S sheep 50 - 60 kg weights were used. Amygdalin and cobalt chloride were given and the eyanide and thioeyanate levels in samples were determined Then some toxicokinetic parameters of the cyanide and thiocyanate in the blood samples taken at 0.5. 1,2. 4. 8, 24. 48 and 72 th hours were established. The cobalt chloride was given intravenously in doses 5 and 1S mgVth hours; the maximal blood levels were respectively, 0.120 and 0128 µg/ml were determined. Besides, area under the curve values according to doses were respectively, 7.09±1.24 and 5.93±0.64 µg. hour/ml; volumes of distribution at steady state were respectively, 0.099±0.005 and 0.125±0.01 1/kg; clearance values were respectively, 1.21±0.44 and 2.52±1.21 m/hour. kg were calculated. The distribution half-Iives of blood thiocyanate levels according to doses were respectively. 0.30±0.02 and 2.49±1.12 hours; the elimination half-lives were respectively, 213.89±11.21 and 96.56±9.35 hours; mean residence times were respectively, 309.09±25.76 and 142.95±15.66 hours; the duration of maximal concentration were respectively, 2 and 1th hours; the maximal blood levels were respectively, 0.028 and 0.032 µg/ml were determined. Besides, area under the curve values according to doses were respectively, 8.77±1.44 and 3.26±1.55 µg. hour/ml; volumes of distribution at steady state were respectively, 0.211±0.01 and 1.333±0.05 1/kg; clearance values were respectively, 0.56±0.23 and 7.66±1.55 ml/hour. kg were established.
